What information is available on drug accumulation with Cialis® (tadalafil) once-daily dosing?
Drug accumulation occurs for up to 5 days after starting tadalafil once-daily dosing. Additional dosing after that time will not result in further accumulation.
Drug Accumulation With Once-Daily Tadalafil
When tadalafil is taken daily, no further accumulation is observed after a certain period of time (steady state).1
In clinical pharmacology trials involving daily doses up to 20 mg in healthy subjects, steady-state plasma concentrations of tadalafil were achieved within 5 days of dosing.1
Tadalafil drug accumulation will occur for up to 5 days, and additional dosing with the same dosage of tadalafil after that time will not result in further accumulation. The degree of drug accumulation after 5 days of tadalafil daily dosing is 1.6-fold greater than that observed after a single dose.1
